Via Buffalo New York is the literal only way, but Northeast Regional to New York then Maple Leaf to Toronto.

It’s possible, just a massive pain because the only train through Indy only comes in the middle of the night 3 days a week but the route is Cardinal to Chicago, then Lakeshore Limited to Buffalo where you transfer to the Maple Leaf to Toronto. It could be good to drive to Chicago then take the train to Chicago from there

You should add a line from Des Moines to Seattle so I don't have to go to Minneapolis (where my ex lives) to join the Greater Seattle polycule.
Take the California Zephyr it stops somewhat close to Des Moines to Emeryville then transfer to the Coast Starlight to get to Seattle.

Where could I take a train to and manage to be back soon? Like, I have 2 days off, but I have to be back at work the afternoon of day 3. Where can I go, assuming the train is the point, not the destination.
(However, getting to do some touristy things would be fun, too)
The best city you can get to is probably Charlotte which takes 5 hours to get to on the Cresent line. The hours are awkward with you arriving in Charlotte at 5 am and leaving at 2 am but it does somewhat work.

Take the Pacific Surfliner to LA then transfer to the Coast Starlight and ride till you hit Emeryville, from there you can be picked up by your gorgeous girlfriend

There used to be a direct route called the Pioneer, but currently you need to take the Coast Starlight to Emeryville and transfer to the California Zephyr to get to Denver
